Overview

Overview

Extracellular Vesicles (EVs) have gained importance since the early 2010s. MSC-derived EVs have been shown to be crucial in wound healing, angiogenesis and immune signaling. However, collection and purification are difficult — undesired exosomes are abundant in most MSC culture media.

DuoEXO is an EV-reduced, xeno-free cell culture supplement for hMSC. Despite the low background noise, MSCs secrete EVs and remain healthy for passages. DuoEXO makes it easier to collect and study MSC-derived EVs with less impurity.

Manufactured under GMP and ISO 13485 certified factory; culture performance is tested batch to batch.

Numbers of nano-particles (diameter 50–200 nm) in basal medium with:

  • B. Brand S, exosome-depleted FBS

The nano-particles in DuoEXO is 100 times smaller than other medium.

Nano-particle counts (50–200 nm) in DuoEXO are roughly 100× lower than basal medium with 10% FBS, exosome-depleted FBS, ultracentrifuged FBS, serum-free brand-G medium, or 5% hPL.

Comparable doubling time to 10% FBS / 5% hPL

UC-MSC doubling time

Doubling time of human umbilical cord-derived MSC (UC-MSC) cultured using either DuoEXO, 10% FBS or 5% hPL as supplement from P1 to P5.

Doubling time of UC-MSC from P1 to P5 cultured in DuoEXO is comparable to 10% FBS or 5% hPL.

Comparison of 5% hPL and DuoEXO: the left panel was medium only, the right panel was medium after MSC culture. Medium was collected and followed by ultracentrifugation and Western Blot. (CD9: exosome marker) The result demonstrates that before culture, unidentified exosomes were detected in 5% hPL but not in DuoEXO. Exosomes were detected in DuoEXO after cultured with MSCs.
